The fluorescence exciting light is fed into the photometer via a light guide. The fluorescence light is detected by a photo-multiplier system. A slit allows both, restricting the exciting light to the preparation and detecting the fluorescence light exclusively from the excited area. A light intensity monitor detects the exciting light intensity. Dividing the fluorescence light signal by the monitor signal deprives fluctuations in the exciting light intensity.
The ultra-fast wavelength changer HSWES changes the fluorescence exciting light from one wavelength wave length to the other within 75µs. A signal sorter unit sorts the fluorescence signal due to the two exciting wavelengths into two DC channels. |
